All of the checkboxes work as expected on first click. Could you please try to reproduce the issue using a new clean Firefox profile (https://goo.gl/AWo6h8), maybe even safe mode (https://goo.gl/AR5o9d), to eliminate custom settings as a possible cause?

I somehow missed your first step, so disregard the new profile request. Could you please post all the information from about:support?

@:cmuresan, You should open about:preferences fist, and then click "Home". If you directly open about:preferences#home, it works as expected. The problem is reproduced not only mouse click but also keypress [Space]. The first keypress [Space] does nothing, it needs keypress [Space] again to toggle checkbox.

This.. seems.. to be fixed in nightly 20180423100754 ?? I'll try to see what changed..

This was fixed by bug 1453227. It even uses Home for a STR in bug 1453227 comment 7 ;)
