I have selected "Check my spelling as I type", but I do not get any spell checking functionality. This has never worked on my PC with either of the betas. However, the functionality does appear to work on my work laptop (Win2k3) in the nightly bon echo builds.

1. Install the beta
2. Make sure that the option to spell check is enabled

The en-GB locale doesn't ship a dictionary because of licensing issues (see bug 321742 for background), and this is release noted at
http://www.mozilla.org/projects/bonecho/releases/2.0b2.html#issues
Eventually the dictionaries will be packaged correctly and available on add-ons.mozilla.org, but in the meantime you have to work around by installing the dict from http://www.mozilla.com/thunderbird/dictionaries.html and then move the files from <app_dir>\components\myspell to <app_dir>\dictionaries. Right click on a text box and use the languages popout to select en-GB.
Most likely spell check works in your nightly because you are using the en-US locale, which does include a dictionary.
